Automated Pipetting: Multiple Dispenses from One Aspiration

Application guide

Multiple dispensing from one aspiration is an automated pipetting task. This page explains component roles, operating sequence, actual operating conditions and restart requirements. Identify the architecture before comparing configurations.

Applicable architecture

This page compares fixed-probe liquid-displacement and disposable-tip air-displacement configurations. Probe or consumable shape alone does not establish the drive architecture. Confirm the actual displacement method and wetted components from the equipment design.[2]

01Where multiple dispensing from one aspiration fits in the instrument

Calculate each delivery, air gap, first and last dose, and retained volume for the actual program. Identify every channel and receiver. Nominal capacity is not all available as dispensable reagent. Compare first, middle, last and first-after-refill doses rather than the average alone.

02Product responsibilities in this operation

The air-displacement pump drives liquid inside the tip; the platform manages tips, wells and movement. Associate liquid destinations and consumable states with every operation.

For this automated pipetting task, define individual dose, total cycle volume, refill or bottle-change interval, backpressure and receiver geometry.

03Organize the sequence by actual architecture

  1. List dose count, individual volume, retained volume and destination-well sequence. Determine the usable total dispense volume within the tip's permitted capacity.
  2. Check pickup sealing and source liquid level, then aspirate once using the actual liquid class. Associate the available volume with the destination-vessel list.
  3. Dispense well by well, recording actual receipt and remaining volume for each dose. Compare first, middle and last doses separately; a cycle average cannot replace individual-dose results.
  4. When aspirating again or changing tips, handle sources and consumables according to the contamination-control strategy and check the next cycle's first dose. After an interruption, identify completed and partially filled wells before choosing where to resume.

04Define operating conditions for this task

Conditions, results and abnormality records for this operation
To defineDetails
Actual operating conditionsDose per operation, total cycle volume, replenishment/bottle-change interval, backpressure and receiving geometry
Acceptance targetUse actual media to check first, middle and last doses and delivery after bottle changes; confirm received volume and timing rather than substituting commanded steps for volume
Fault localizationEmpty sources, bubbles, nozzle droplets and backpressure changes affect first or last doses

06Validate operation results at the receiver

Use actual media to check first, middle and last doses and delivery after bottle changes; confirm received volume and timing, not commanded steps as a proxy for volume.

Measure delivery with actual liquids, tips and well positions, including idle periods, refills and multiple dispensing.

07Locate abnormalities and restore the next operation

Empty sources, bubbles, nozzle droplets and backpressure changes may affect first or last doses.

Stop affected steps, validate recovery after priming and avoid duplicate dispensing.

Nominal capacity and volume per step do not establish the minimum reliable pipetting volume. Disposable tips and reusable probes require different cleaning strategies.

What is a programmable automatic air-displacement pipetting pump?

A programmable automatic air-displacement pipetting pump is a liquid-handling component whose aspiration, dispense, speeds and sequence are set through control commands. This article focuses on air-displacement modules that use disposable tips in OEM instruments, with FOREACH SMTP2 as an example of operation, monitoring and selection limits.

What Is Pipetting? Manual and Automated Workflows Compared

Pipetting is the transfer of a specified volume of liquid from one location to another. Examples include moving 100 µL from a sample tube into a reaction vessel, or adding sample and diluent in a defined ratio.
