Nucleic acid processing: washing and elution

Application guide

This guide covers washing and elution within nucleic-acid processing: component roles, action sequence, operating conditions and restart requirements. Identify the architecture before comparing configurations.

Applicable architecture

This page covers liquid handling in nucleic acid processing. For magnetic-bead extraction, first establish whether the architecture transfers liquid or beads. Pumps serve only the liquid operations actually required; separation steps do not all involve pumping through fixed tubing.[2]

01Where washing and elution occur in the instrument

Identify the separation architecture first. When pipetting liquid while retaining the solid phase, plan supernatant removal, residual-wash removal, elution and product collection separately. When magnetic rods move beads, organize actions around reagent wells and solid-phase movement. For liquid-driven steps, check aspiration height, the complete wetted path and actual product receipt.

02Product responsibilities in this operation

The air-displacement pump drives liquid inside the tip; the platform manages tips, wells and movement. Associate liquid destinations and consumable states with every operation.

For this nucleic acid processing task, define solid-phase position and retention target, wash cycles, aspiration height, residual liquid, destinations and downstream method requirements.

03Organize the sequence by actual architecture

  1. Confirm a tip-based liquid-transfer architecture that retains the solid phase, and mark separate destinations for wash waste and eluate. Organize magnetic-rod bead transfers around the corresponding solid-phase movements.
  2. Set aspiration height from solid-phase position and vessel geometry. Remove supernatant, add wash liquid and remove residual wash liquid separately, without discarding the solid phase that must be retained.
  3. Complete elution and the required solid-phase separation according to the method, then use a tip to collect the retained product into the designated vessel.
  4. Record actual product volume, target recovery, residual wash liquid and contamination blanks. Evaluate the workflow through these results and downstream usability; completion of pipetting alone is insufficient.

06Validate operation results at the receiver

Evaluate solid-phase loss, post-wash residue and downstream assay background or nucleic-acid recovery together.

Record product volume, recovery and downstream results, and check sample-to-sample contamination.

07Locate abnormalities and restore the next operation

Insufficient capture, incorrect aspiration height, uneven coverage or pooling may impair washing and recovery.

Review actual results of affected steps before resuming the next action.

Extraction and library-preparation liquid handling differ in contamination and outcome requirements. Confirm research workflows and clinical diagnostic methods separately.

What is a programmable automatic air-displacement pipetting pump?

A programmable automatic air-displacement pipetting pump is a liquid-handling component whose aspiration, dispense, speeds and sequence are set through control commands. What Is Pipetting? Manual and Automated Workflows Compared

Pipetting is the transfer of a specified volume of liquid from one location to another. Examples include moving 100 µL from a sample tube into a reaction vessel, or adding sample and diluent in a defined ratio.
