Where online sampling occurs in the instrument
Define what the online sample measures, its destination and whether sample discard is allowed. Assess sampling for representativeness, path retention, contamination and disturbance to reactor volume, linked to the main bioprocess task.
The stagnant section between the sampling port and receiver needs refreshing, but discarded liquid consumes process volume. Define the analytical objective and sampling budget together.
Product responsibilities in this operation
Syringe drive and valve positions jointly organize aspiration, holding and dispensing. Record working volume, remaining inventory and replenishment timing according to the sequence.
For this process feeding and sampling task, define the analyte, sampling interval, stagnant-volume refresh, discarded-sample budget and permitted pretreatment.
Organize the sequence by actual architecture
- Define the analyte, sampling location, receiving destination and permissible discard volume. Include allowable disturbance to the reaction-vessel volume in the budget.
- Refresh the actual stagnant path and record discarded refresh liquid separately from the collected sample. Pump stroke alone does not establish that the sample has been refreshed.
- Deliver the sample to its specified receiver. Check refresh time and analyte representativeness, and reconcile process volume after sampling, sample discard and other drainage.
- Complete the required path isolation or maintenance and record contamination status. After interruption, check the amount already withdrawn and current destination before defining the restart sequence.
Define operating conditions for this task
| To define | Details |
|---|---|
| Actual operating conditions | Analyte, sampling interval, renewal of stagnant sections, discard-volume budget and permitted pretreatment |
| Acceptance target | Measure renewal time at the analytical or receiving interface and assess target representativeness using paired samples |
| Fault localization | Old-sample mixing, settling, adsorption or insufficient renewal time can make the sample unrepresentative of its source. |
Compare product configurations by the roles on this page
| Product | Configuration option | Conditions to verify |
|---|---|---|
| HLD rotary-valve syringe pump | Aspiration, holding and dispensing with a rotary valve | Check source/outlet valve positions, capacity budget and replenishment timing |
| HMD solenoid-valve syringe pump | Metered aspiration and dispensing with solenoid valves | Check open/closed and power-off states, working volume and replenishment cycle |
Validate operation results at the receiver
Measure renewal time at the analytical or receiving interface and assess target representativeness using paired samples.
Connect the culture process, actual feeding and sample analysis, retaining records across the entire run.
Locate abnormalities and restore the next operation
Old-sample mixing, settling, adsorption or insufficient refresh time may make the received sample differ from its source.
Review actual results of affected steps before resuming the next action.
Periodic dispensing, continuous feeding and aseptic sampling have different requirements. A single metering test cannot establish all three.
