Spectroscopy and elemental analysis: standards and dilution

Application guide

Standards and dilution are specific liquid-handling operations in spectroscopy and elemental analysis. This page explains component roles, action sequence, actual operating conditions and recovery requirements. Establish the architecture before comparing configurations.

Applicable architecture

This guide covers liquid-sample elemental analysis and auxiliary fluidics. Solid, gas and other spectroscopic architectures require separate assessment of their introduction devices. Flow or capacity alone does not qualify these candidates as replacements for the original sample-introduction system.

01Where standards and dilution occur in the instrument

Provide separate paths as needed for standards, over-range dilution, internal standards and continuous auxiliary reagents. Verify concentration and mixing from actual delivered sample and reagent volumes. The method determines whether continuous supply is required.

Measure delivery of stock, diluent and internal standard separately, then validate the resulting concentration or method response after mixing. Shared-section residue can produce an incorrect composition despite correct commanded volumes.

02Product responsibilities in this operation

Syringe drive and valve positions jointly organize aspiration, holding and dispensing. Record working volume, remaining inventory and replenishment timing according to the sequence.

For spectroscopy and elemental analysis, confirm actual sample, diluent or internal-standard volumes, concentrations at each stage, mixing and secondary-sampling conditions.

03Organize the sequence by actual architecture

  1. List stock, diluent and internal-standard sources, target concentrations and working volumes according to the method. Specify receiving locations and mixing methods.
  2. Deliver each liquid through the source-selection and syringe aspiration/dispense sequence. Record actual receipt and the effect of shared-path flushing on composition.
  3. After mixing, check actual ratio, concentration or method response, including low-range preparation, overrange dilution and the first liquid after source changes.
  4. Confirm washing and blank recovery for the actual matrix. If the method requires continuous internal-standard or auxiliary-liquid addition, configure that branch separately for continuous flow and merging conditions.

04Define operating conditions for this task

Conditions, results and abnormality records for this operation
To defineDetails
Actual operating conditionsActual volumes of original sample, diluent or internal standard; concentrations at each stage; mixing and resampling conditions
Acceptance targetMeasure each branch's delivery, received ratio and mixing uniformity separately, then check assay response
Fault localizationSmall-volume errors, shared residual liquid or inadequate mixing may alter the actual ratio

06Validate operation results at the receiver

Measure each branch's delivery, received ratio and mixing uniformity separately, then check assay response.

Check concentration ratios, mixing, blank recovery and method results. Evaluate preparation and sample supply separately.

07Locate abnormalities and restore the next operation

Small-volume errors, shared residue or insufficient mixing may change the actual ratio.

Review actual results of affected steps before resuming the next action.

A sample-preparation pump is not automatically suitable for nebulizer supply. Confirm the introduction system for the specific instrument.

What Is a Programmable Syringe Pump? Operation, Fluid Routing and Automation

A programmable syringe pump uses a motor to control syringe plunger displacement and speed, executing aspiration, dispensing, pauses and repeated actions through commands. With a routing valve, it can connect reagent reservoirs, reaction vessels and wash paths into an automated dispensing process. Instrument development therefore needs to define volume, fluid connections and timing together.

What Is Pipetting? Manual and Automated Workflows Compared

Pipetting is the transfer of a specified volume of liquid from one location to another. Examples include moving 100 µL from a sample tube into a reaction vessel, or adding sample and diluent in a defined ratio.
