Tubing and fittings in microplate dispensing and washing

Application guide

This page explains tubing and fitting locations, coordination and configuration requirements for reservoir, manifold and waste connections in microplate dispensing and washing, with links to relevant operating and validation tasks.

Applicable architecture

Solid-phase retention and washing conditions apply to microplate methods that include those tasks. Configure simple dispensing or other plate operations for the actual station. Valve port count is not the number of channels that can independently supply liquid simultaneously.

01What do these products do in this instrument?

Tubing and fittings: connect reservoirs, manifolds and waste paths

Build supply and aspiration networks for the plate station, identifying shared sections and independent branches. Include length, bore and interface differences in per-well or per-channel delivery checks.

Tubing and fittings together form sealed connections and the actual transfer path. Check formulation, inner/outer diameters, length, tube-end positioning and port geometry, including added volume in residue and arrival delay.

02Design tubing and fittings as a complete wetted path

Tubing, fittings and ports jointly determine sealing, transfer volume and medium-contact coverage. Materials and dimensions must suit formulation, operating conditions and installation. Include tubing length and interface volume in priming, shared residue and arrival delay. After connection changes, recheck delivery and cleaning through the actual path.

03Calculate connection volume and post-maintenance recovery

Shared volume affects liquid arrival and flushing; interface gaps may retain the previous composition. Record length, actual bore and tube insertion position. After disassembly, reprime and check seals, then compare first-cycle delivery, blanks or recovered product to confirm path recovery.

05Manifold connections

Assess flow distribution and air aspiration against branch length, bore size, fittings and manifold volume. After assembly, check sealing, blocked ports, flushing and maintenance access before measuring well-to-well performance.

After assembly, check blocked ports, leaks and path volume, and measure each well. Calibrating total volume alone can conceal local dispensing errors.

Check leaks, tube disconnection and material changes at actual pressure and temperature; evaluate residue, recovery and the first post-maintenance cycle through the complete path

Misconnections, gaps, tube deformation or leaks may alter volume and pressure. Isolate the affected segment and reconfirm assembly; matching threads do not establish interchangeability.

07Related operations and architecture comparisons

Microplate washing and aspiration

Observe residual liquid, foam and solid-phase loss during each cycle. Insufficient supply, a misaligned wash head and delayed waste removal can cause similar symptoms and must be checked separately.

Manifold connectionsCheck leaks, tube disconnection and material changes at actual pressure and temperature; evaluate residue, recovery and the first post-maintenance cycle through the complete pathMisconnections, gaps, tube deformation or leaks may alter volume and pressure

Assess differences in delivered volume between wells, residual liquid after washing and the complete plate cycle. Preserve well identities.

09Does a fitting that tightens prove the tubing assembly is suitable?

Also confirm port geometry, tube-end positioning and seal fit. Check leakage, hold-up and performance after repeated maintenance under actual conditions; thread engagement alone is insufficient.

Multiport selection does not provide independent simultaneous channels. Validate washing through both liquid handling and retention of the required solid phase.

What is flow resistance calculation for? Using the Foreach calculator in fluid-path design and selection

The free flow rate of the pump meets the requirements. After installing the pipelines, valves and joints, the liquid supply at the end is insufficient. Flow resistance calculations can break the problem down into comparable data: what pressure differential is required at the target flow rate, where does the pressure loss mainly occur, and what happens if the pipe diameter, pipe length, component Cv or medium viscosity is changed. This article combines the actual function of the Foreach flow resistance calculator with a recalculable fluid circuit case to explain how to use the calculation results for design, selection, troubleshooting and prototype verification.

Why Does Actual Flow Change So Much When the Tubing Size Changes on the Same Miniature Diaphragm Pump?

Even when the pump is unchanged, installed flow can change because of tubing inner diameter, length, bends, and the minimum bore of fittings. When flow falls after a tubing change, the effective response is neither to blame the pump first nor to enlarge every tube indiscriminately. Measure inlet pressure, outlet pressure, and accumulated flow together to determine where the added restriction actually occurs.

Why do multiple wash nozzles deliver different flows from one miniature diaphragm pump?

Calculate demand from the volume and timing of each wash nozzle, separate insufficient pump output from unequal distribution, and verify each branch with collection and pressure measurements.

Discuss a microplate workstation fluidic configuration

Provide a fluid diagram, the actual liquids and pressures in each state. Bore size, length, load and actual formulation jointly determine material and dimensional choices. Check tubing OD, threads, sealing structure and port-bottom geometry. Compare suitable configurations against the validation objectives described here.
