Tip-Based Dilution and Dispensing: Linking Ratios to Actual Delivery

Application guide

Tip-based dilution involves sample, diluent, mixing and destination wells. Multiple dispenses from one aspiration also require the identity of each dose; total volume in the pump chamber alone is insufficient.

01Locate the source, drive and receiving position for this task

Original sample and diluent→Tip-based metering→Mixing in the target well or vessel→Subsequent sampling
Functional or operational sequence; confirm connections and branches for each state on the instrument

Define actual sample, diluent or internal-standard volumes, concentration at each stage, mixing and subsequent sampling conditions. Follow the diagram to confirm liquid contact, pressure and destination at each position before assigning drive and connection roles.

02Calculate cycle volume from dilution ratio and well count

Calculate sample, diluent and total cycle volume from the ratio and well count, including first and last doses and retained volume. Small sample aliquots may need a different tip or program. Adequate capacity does not establish acceptable ratio error.

03Compare product options against task requirements

The following candidate options match this page's responsibilities. Compare them against the operating conditions described, then check capacity, materials and interfaces on the product pages. Card images show representative configurations; suitability must still be confirmed at actual operating points and by application results.

04Preserve identity through mixing, tip changes and sampling at each level

Perform sampling, diluent addition and mixing in sequence, changing tips according to contamination controls. Record each source in serial dilution. Preserve completed-well records after replenishment or faults to prevent duplicate dosing.

Action records should link commands, valve or interface states and actual received results. When raw material, container or sample identity changes, reconfirm shared paths and conditions permitting the next step. Completion of drive movement alone does not replace task acceptance.

05Include minimum sample aliquots and first and last wells

Measure sample and diluent delivery first, then validate mixture ratio and method results. Include first and last wells, minimum sample aliquots, replenishment and different liquid classes.

Link operating-point tests to interpretable results
Validation scenarioMeasurement or observationInterpretation and action
Minimum sample volumeMeasure delivered volumes and mixed ratio separatelyCheck tip and air-column condition when small-volume delivery fails
First and last wells and serial stagesCover first and last wells and subsequent samplingDistinguish dose, mixing and propagated stage-to-stage errors

Record test medium, configuration, installed path and operating sequence; present routine operating points and boundary results separately. Calculations support design discussions, component documentation checks models, and installed measurements confirm the actual task.

06After an abnormality or maintenance, first identify which segment needs recovery

Small-volume error, shared-path residue or insufficient mixing can change the actual ratio. Check results from the affected step before resuming the next action.

Retain current source, container identity, completed actions, valve or interface state and actual receiving information. Use the table to locate the stage that changed before deciding to retry, continue or restart. After recovery, recheck the relevant operating point before returning an uncertain action to the automatic cycle.

What Is Pipetting? Manual and Automated Workflows Compared

Pipetting is the transfer of a specified volume of liquid from one location to another. Examples include moving 100 µL from a sample tube into a reaction vessel, or adding sample and diluent in a defined ratio.
